Instructions for In-Pouch™
- Sample Collection
- Materials needed (per animal):
- InPouch™ TF
- Disposable gloves
- Infusion/insemination pipette/Pizzle Stick (Lane Mfg)
- 20mL syringe/pipette bulb
- Collect the sample:
- Bulls: The pipette or scraper should be inserted into the preputial orifice with caution to not allow any outside debris or feces to touch the sampling device. The pipette is then directed caudally toward the preputial fornix (10-12 inches). Smegma should be collected by the device using a combination of scraping and aspiration with an attached disposable syringe.
- Cows: Advance the pipette gently to the floor of the vaginal fornix, and aspirate mucus with a syringe.
- Innoculation
- Remove pouch from the box, and manually express approximately 0.5 mL of liquid to the top chamber. Open the pouch by tearing off the top along the scored line. Use the white tabs to open the mouth of the pouch.
- Insert the sample into the upper chamber of the pouch using the fluid within the top chamber to flush smegma from collection device.
- Gently push the entire sample into the bottom chamber. Avoid the formation of bubbles.
- To close the pouch, roll the top down tightly until the wire tape is at the top of the label. Fold the wire tape ends to seal the pouch. Avoid trapping air. Complete the information on the label, ensuring a permanent ID for the bull or cow is used.
- Following inoculation of pouch, store the pouch upright at room temperature and away from light.
- Submission
- Place sealed InPouch™ TF into provided plastic bag
- Affix shipping label on outside of original kit box
- Include the completed ADDL Ruminant Submission Form
- Culture – 6 day turn-around time
- PCR – 2-3 day turn-around time
- Ship overnight to the ADDL (or hand deliver) with no ice pack to the West Lafayette location
